Năm 2016 là năm đầu tiên chương trình áp dụng luật xuất phát cách nhau 5 phút. Thứ tự xuất phát của các đội đua ở chặng sau sẽ dựa vào thứ tự về đích ở chặng trước đó. Đội về đích thứ nhất sẽ xuất phát trước đội thứ hai đúng 5 phút, đội về thứ 2 sẽ xuất phát trước đội về đích thứ ba đúng 5 phút và cứ như vậy áp dụng lần lượt cho các đội còn lại. Với luật chơi 5 phút, dù đội về trước cách đội về sau khoảng thời gian bao lâu chăng nữa thì sang chặng tiếp theo, họ sẽ xuất phát trước đúng 5 phút. Nếu như ở những mùa đua trước, đội về nhất cách đội về nhì 2 tiếng, đến chặng sau, họ xuất phát trước đội về nhì 2 tiếng thì với luật 5 phút của Cuộc đua kỳ thú năm nay, đội về nhất cũng cách đội về nhì 5 phút. Với luật chơi mới này, các đội đua bắt buộc phải cố gắng, nỗ lực hết mình trong tất cả các chặng.
